Determination of effective compounds in ethanolic extract of propolis (Kurdistan) by High Performance Liquid Chromatography (HPLC)

Background and Aim: Propolis, as a natural product, has been shown to provide several biological benefits including antioxidant, anti-inflammatory, and antimicrobial as well as anti-cancer properties. Its application extensively depends on its composition and the amounts of the major active components. Therefore, in the presented study we aimed to evaluate the amounts of chrysin, quercetin and, caffeic acid as three active components, in the Iranian propolis using the UV-HPLC method.
 
Materials and Methods: Three different samples of propolis were collected from Saral & Avalan regions (Kurdistan Province, Iran) during spring 2017. To isolate and measure the three major propolis compounds, the samples were extracted with an ethanolic solution (80%) and then injected into the UV-HPLC system with an isocratic mode at a flow rate of 1ml/min for 20 min and ultraviolet detection at 260 or 310 nm. The linearity and detection limits of the method were calculated to evaluate the quality control of the assay.
 
Results: The average amounts of quercetin and caffeic acid were 65.425 and 27.498 µg/g, respectively. While chrysin was not found in propolis samples.
 
Conclusion: The results of this study showed that ethanolic extract of propolis collected from Kurdistan contains two main flavonoids quercetin and caffeic acid (compounds rich in natural antioxidants).
