Evaluation of the effect of nicotinic acid and folic acid on morphology, membrane integrity and quality of sperm chromatin concentration in normozoospermic men during cryopreservation.

Introduction: Cryopreservation of sperm is an Assisted reproductive technique, but with[سارا1]  production of oxidative stress, sperm function is disrupted. This study was carried out to evaluate efficacy of Nicotinic acid and Folic acid during [DD2] cryopreservation on sperm parameters in order to preserve sperm functions.
Methods:30 semen samples were obtained from  normozoospermia men,aged between 20 and 45 years old.Each sample was evaluated from the point of quality of chromatin,viability,membrane integrity,morphology and motilityof sperms and divided into four groups for freezing:Control,Folic acid(50 nM),Nicotinic acid(10mM) and a combination of nicotinic acid (10 mm) + Folic acid (50 nM).After freezing, the parameters were evaluated again, and in each group compared with before freezing with other groups.Quality[سارا3] [DD4]  of chromatin with Toluidine blue staining,viability by Eosin-Nigrosin staining,membrane integrityby HOST method and morphology and mobility evaluated by computer-aided sperm analysis (CASA) respectively.
 
Results: The quality of chromatin, viability, membrane integrity, normal morphology and sperm motility of rapid slow, and non-progressive was less and immotIle sperms was more in all cryopreserved groups than before freezing group (p                                                                                                                                                                                                                 
Conclusion: Nicotinic acid and folic acid during cryopreservation exhibited positive effects by enhancing sperm quality.
