Tetrandrine, an Immunosuppressive Alkaloid, Reduces TNF-α Secretion in Adipose Tissue–Derived Mesenchymal Stem Cells (AD-MSC).

Background and Aim: Mesenchymal stem cells (MSCs) have shown different immunomodulatory properties in the treatment of various inflammatory and autoimmune diseases due to cell-to-cell contact, as well as the secretion of some soluble factors. In order to promotion the therapeutic activities of these cells, researchers have suggested preconditioning them with various components, including natural extracts. This study aimed to evaluate the effect of tetrandrine on the secretion of TNF-a and IL-6 cytokines from adipose tissue-derived mesenchymal stem cells (AD-MSCs).
Materials and Methods: The levels of TNF-a and IL-6 in the supernatant of tetrandrine-treated AD-MSC cells were measured by ELISA method after 48 hours.
Results: Our data showed that treatment of AD-MSCs with selected doses of tetrandrine could reduce the level of TNF-a as an inflammatory cytokine in MSCs (P < 0.01), but no significant change was observed in the level of IL-6 compared to the control group.
Conclusion: Our results showed that tetrandrine increases the anti-inflammatory potential of MSCs. Hence, it is possible that the treatment of MSCs with tetrandrine enhances the therapeutic and immunomodulatory properties of these cells.
