Background and Aim:Mesenchymal stem cells (MSCs) have shown different immunomodulatory properties. They suppress the proliferation and activation of T and B lymphocytes, inhibit natural killer cell activity, and impair dendritic cell maturation. In order to promote the therapeutic activities of these cells, researchers have suggested preconditioning them with various components, including natural extracts. Tetrandrine is a bisbenzylisoquinoline alkaloid with well-documented immunosuppressive and anti-inflammatory properties that modulates cytokine production and immune cell functions. This study aimed to evaluate the effect of tetrandrine on the secretion of TNF-a and IL-6 cytokines from MSCs. Materials and Methods: MSCs were cultured and treated with tetrandrine for 48 hours, after which the levels of IL-6 and TNF-a in the culture supernatant were measured using a sandwich enzyme-linked immunosorbent assay (ELISA). Results:The findingsshowed that the treatment of AD-MSCs with various doses of tetrandrine could reduce the level of TNF-a as an inflammatory cytokine in MSCs (P < 0.01); however, no significant changes were observed in the level of IL-6, compared to the control group. Conclusion:According to theresults, tetrandrine increases the anti-inflammatory potential of MSCs through decreased secretion of TNF-a. Hence, it is possible that the treatment of MSCs with tetrandrine enhances the therapeutic and immunomodulatory properties of these cells.
