Background and purpose: The aim of this study was to investigate the effect of cerium oxide nanoparticles on the expression of some genes related to apoptosis and testicular development in mouse embryos. Materials and methods: CNPs were injected intraperitoneally at 25, 80, and 250 mg/kg concentrations on days 7 and 14 of gestation. At the end of the pregnancy, the neonates testicles were destroyed on the sixth day after birth. Real time-PCR was used to study Gata4, Sox8, Rad54, Bax, Bcl2, Caspase3, Caspase8 and Gsk3-β genes. Results: The expression of Gata4, Sox8, and Rad54, Bcl2, and Gsk3- genes was significantly reduced at concentrations of 80 and 250 mg/kg. At the same concentrations, the Bax, Caspase3, and Caspase8 genes showed a significant increase in expression. Changes in the expression of all studied genes at a concentration of 25 mg/kg were not statistically significant, Furthermore, at concentrations of 80 and 250 mg/kg, the expression ratio of Bax/Bcl2 genes was significantly higher, and this ratio did not change significantly in the 25 mg group. Conclusion: According to the findings of this study, cerium oxide at concentrations of 80 and 250, in addition to inducing apoptosis in neonatal mouse testicular cells, influences the development of this organ by influencing the expression of genes involved in testicular development. However, because the concentration of 25 mg of this nanoparticle had no significant effects on the testicular development process of newborns, this concentration may be used in different cases with further research.
