The effect of Oleoylethanolamide supplementation on prostaglandin concentration and pain intensity in primary dysmenorrhea; A Randomized, double-blind, controlled clinical trial

Background and Aim: Primary dysmenorrhea is a common gynecological health disorder. The aim of this study was to determine the effects of oleoylethanolamide (OEA) supplementation on pain intensity and inflammatory prostaglandins concentration in girls with primary dysmenorrhea.
Materials and Methods: This double-blind clinical trial was performed at Qazvin University of Medical Sciences in 2020 in which 44 girls with primary dysmenorrhea were divided into two equal groups by simple random sampling method and received one 125 mg OEA capsule in the intervention group and 125 mg capsule containing wheat flour in placebo group daily for 8 weeks. After collecting demographic information, at the beginning and end of the study, the questionnaires of physical activity, 24-hour food recall, and visual analog scale were completed and prostaglandin concentrations E2 and F2α were measured. Paired sample t-test was used to analyze the changes in each group at the beginning and end of the study and an independent t-test was used to analyze the differences between the two groups.
Results: At the beginning and end of the study, there was no significant difference between the two groups in terms of anthropometric factors, food intake, and physical activity (P> 0.05). At the end of the study, consumption of OEA significantly reduced the mean pain intensity (from 7.01±2.54 to 5.30±2.32), the concentration of prostaglandins E2 (from 1236.65±404.37 pg/ml to 918.58±276.42 pg/ml) and F2α (from 2073.65±706.31 pg/ml to 1611.65±598.84 pg/ml) compared to the beginning (P <0.05). No significant change was observed in pain intensity and prostaglandin concentration in the placebo group (P> 0.05).
Conclusion: Given that OEA supplementation reduces the concentration of prostaglandins and the severity of dysmenorrhea pain, use of this supplement can be introduced as an alternative to reduce the use of anti-inflammatory drugs.
