fa بررسی خواص آنتی باکتریال عصاره الکلی پروپولیس کردستان بر روی پسودوموناس آئروژینوزا، باسیلوس سرئوس و استافیلوکوکوس اورئوس Antimicrobial activity of aqueous and alcoholic extracts of Kurdistan propolis on Staphylococcus aureus, Bacillus cereus and Pseudomonas aeruginosa عمومى General پژوهشي اصیل Original Research مقدمه: باکتری های استافیلوکوکوس اورئوس باسیلوس سرئوس و سودوموناس آئروژینوزا به طور معمول میتوانند مسمومیت های غذایی و بیماری های گوارشی را در انسان ایجاد کنند. عصاره بره موم (propolis) به عنوان ترکیب ضد میکروبی در مقابل باکتری ها شناخته شده است. هدف از این تحقیق تعیین فعالیت ضد میکروبی عصاره بره موم  بر روی سویه های باکتریایی در شرایط آزمایشگاهی می باشد. روش بررسی: در این تحقیق، اثرات ضد باکتریایی عصاره الکلی بره موم، بر روی باکتریهای استافیلوکوکوس اورئوس (ptcc1112) ، باسیلوس سرئوس(ptcc1247) و سودوموناس آئروژینوزا (ptcc1707) در سه تکرار، به روش دیسک کاغذی (تعیین قطر هاله عدم رشد) و رقت سازی در لوله، تعیین حداقل غلظت ممانعت کنندگی (MIC) و حداقل غلظت کشندگی (MBC) مورد ارزیابی قرار گرفت. جهت آنالیز آماری از نرم افزار SPSS و آزمون آماریDUNCAN  استفاده شد. یافته ها: بر اساس نتایج، فعالیت ضد میکروبی، قطر هاله عدم رشد میکروارگانیسم های های استافیلوکوکوس اورئوس، باسیلوس سرئوس و سودوموناس آئروژینوزا (در غلظت mg/ml 25/5( به ترتیب 58/0±66/18، 18و 58/0±33/14، تعیین شد، که استافیلوکوکوس اورئوس، بیشترین حساسیت و سودوموناس آئروژینوزا بیشترین مقاومت را به عصاره الکلی بره موم نشان داد. نتایج حداقل غلظت ممانعت کننده (MIC) برای های استافیلوکوکوس اورئوس، باسیلوس سرئوس و سودوموناس آئروژینوزا به ترتیب mg/ml 656/0، 656/0و 62/2 تعیین شد و همچنین نتایج غلظت کشنده (MBC) برای کلوستریدیوم اسپوروژنس، اشریشیا کلی و استافیلوکوکوس اورئوس به ترتیب 565/0، 656/0 و62/2 میلی گرم بر میلی لیتر تعیین گردید. نتیجه گیری: عصاره الکلی بره موم بر روی باکتری های استافیلوکوکوس اورئوس، باسیلوس سرئوس و سودوموناس آئروژینوزا نه تنها خاصیت بازدارندگی بلکه خاصیت ضد باکتریایی نیز داشت. با مطالعه اثرات ارگانولپتیک عصاره بره موم در غذا می توان به عنوان یک ماده محافظت کننده استفاده نمود. کلمات کلیدی: بره موم، هاله عدم رشد، ضد باکتری، استافیلوکوکوس اروئوس، باسیلوس سرئوس، سودوموناس آئروژینوزا. <span dir="RTL" lang="AR-SA" style="line-height: 115%; font-family: "B Zar"; font-size: 12pt; mso-bidi-language: AR-SA; mso-fareast-font-family: Calibri; mso-ansi-language: EN-US; mso-fareast-language: EN-US; mso-ascii-font-family: Calibri; mso-hansi-font-family: Calibri;">وصول مقاله:<span lang="AR-SA" style="line-height: 115%; font-family: "Calibri","sans-serif"; font-size: 12pt; mso-bidi-language: AR-SA; mso-fareast-font-family: Calibri; mso-ansi-language: EN-US; mso-fareast-language: EN-US; mso-bidi-font-family: "B Zar";"> <span dir="RTL" lang="AR-SA" style="line-height: 115%; font-family: "B Zar"; font-size: 12pt; mso-bidi-language: AR-SA; mso-fareast-font-family: Calibri; mso-ansi-language: EN-US; mso-fareast-language: EN-US; mso-ascii-font-family: Calibri; mso-hansi-font-family: Calibri;">5/10/94 اصلاحیه نهایی:19/10/94 پذیرش:<span dir="RTL" lang="FA" style="line-height: 115%; font-family: "B Zar"; font-size: 12pt; mso-bidi-language: FA; mso-fareast-font-family: Calibri; mso-ansi-language: EN-US; mso-fareast-language: EN-US; mso-ascii-font-family: Calibri; mso-hansi-font-family: Calibri;">01<span dir="RTL" lang="AR-SA" style="line-height: 115%; font-family: "B Zar"; font-size: 12pt; mso-bidi-language: AR-SA; mso-fareast-font-family: Calibri; mso-ansi-language: EN-US; mso-fareast-language: EN-US; mso-ascii-font-family: Calibri; mso-hansi-font-family: Calibri;">/11/94 Background and Aim: Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus cereus can cause food poisoning and digestive disorders in human beings. Propolis has been known as an antimicrobial agent against bacteria. The aim of this study was to determine the in-vitro antimicrobial activity of propolis extract on bacterial strains. Material and Methods: In this study, we evaluated antibacterial effects of propolis alcoholic extract on Staphylococcus aureus (ptcc1112), Bacillus cereus (ptcc1247) and Pseudomonas aeruginosa (ptcc1707) for three times, by means of paper disc method (determination of inhibition zone diameter) and broth dilution, determination of minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Using SPSS software, statistical analysis was performed by DUNCAN test. Results: The results showed that the antibacterial activity and inhibition diameters for Staphylococcus aureus, Bacillus cereus and Pseudomonas aeruginosa (in a propolis extract concentration of 5.25 mg / ml) were respectively 18.66±0.58, 18, 14.33±0.58, respectively. S. Aureus had the highest sensitivity and Pseudomonas aeruginosa showed the highest resistance to alcoholic extract of propolis. The minimum inhibitory concentrations (MIC) for Staphylococcus aureus, Pseudomonas aeruginosa and Bacillus cereus were 0.656, 0.656 and 2.62 mg/ ml respectively and the lethal concentrations (MBC) for Clostridium sporogenes, Escherichia coli and Staphylococcus aureus, were 0.656, 0.656 and 2.62 mg/ ml respectively. Conclusion: The alcoholic extract of propolis had inhibitory and antibacterial effects on Staphylococcus aureus, Pseudomonas aeruginosa and Bacillus cereus. Considering organoleptic effects of propolis extract, it can be used as a preservative in food.
