بررسی کمی تعداد ماستوسیتهای غده سمینال وزیکول موش صحرایی بالغ در دوره حاد پس ازقطع نخاع
Quntitative Analysis of Mast Cells in Seminal Vesicles of Adult Rat During Acute Phase of Spinal Cord Injury

چکیده
مقدمه: این تحقیق به منظور بررسی تغییرات توزیع ماستوسیتهای غده سمینال وزیکول موش صحرایی در طی دوران حاد آسیب نخاعیSCI (spinal cord injury) صورت گرفت.
مواد و روشها: این بررسی یک مطالعه RCT است که در آن 43 سر موش صحرائی نر از نژاد Sprague دو ماهه به طور تصادفی به دو گروه آزمایش و کنترل تقسیم شدند. نخاع موشهای صحرایی گروه آزمایش پس از بیحسی ولامینکتومی درناحیهT9، بطور عرضی قطع شد. در گروه کنترل تنها عمل لامینکتومی در ناحیه T9صورت گرفت ولی نخاع سالم باقی ماند. در هفته اول و دوم و سوم پس از جراحی ، از غده سمینال وزیکول گروههای کنترل و آزمایش نمونه گیری به عمل آمد و از آنها برشهای سریال تهیه شد و پس از رنگ آمیزی تولوئیدین - بلو تعداد ماستوسیتهای همبندی و مخاطی گروه کنترل و آزمایش محاسبه و با استفاده از تست من ویتنی یو (Mann-Whitney U )معنی دار بودن اختلاف گروههای کنترل و آزمایش بررسی شد.
نتایج: تعداد ماستوسیتهای غده سمینال وزیکول درگروههای آزمایش افزایش یافت. بطوریکه میانگین این تعداد در گروه کنترل 7 روزه 014/8 (22/0 ) و در گروه آزمایش 63/11 (59/0 )، همچنین در گروه کنترل 14 روزه 14/8 (85/0 ) و در گروه آزمایش 7/70 (84/7 ) بود که از نظر آماری نیز معنی دار است (001/0P< ). این افزایش در گروههای هفت روزه و چهارده روزه در مقایسه با گروه کنترل معنی دار بود.
نتیجه گیری: افزایش ماستوسیتها، بویژه ماستوسیتهای مخاطی گروه آزمایش نشان دهنده آسیب اپی تلیومی در این دوران است و ممکن است یکی از دلایل افزایش تیتر آنتی بادی در مایع منی و کاهش کیفیت مایع منی و متعاقبا" ناباروری بیماران نخاعی باشد.
ABSTRACT
INTRODUCTION: This study was aimed to evaluate changes in number and distribution of mast cells in seminal vesicles of rat during acute phase of spinal cord injury (SCI).
MATERIALS & METHODS: This research was a randomized clinical trial. Forty three 2- month-old rats of sprague race were selected and divided to the control and case groups. In the case group spinal cord disscection at the level of T9 was performed following local anesthesia and laminectomy. In the cotrol group, only laminectomy of the T9 was performed but the cord remained intact. In both groups biopsy from seminal vesicls was perfomed in the first, second and third weeks after surgery. Serial sections of speciments were stained with Toluidine-blue. Then mast cells in the connective and mucusal tissues were counted. The results were analysed by mann whitney U test.
RESULTS: The number of mast cells in the case group was increased as compared with control group both in the first (11.630.59 versus 8.0140.22) and second 8.140.85 versus 70.77.84) weeks (P<0.001).
CONCLUSIONS: Increased number of mast cells, particularly mucusal mast cells, is due to epithelial damage during actue phase of SCI. This may be a cause of increase in antibody titer in the semen that may have a roll in infertility of the patients with SCI.
